I am in the process of getting some baits together to sell on here and I get the main concept but what I am wondering is what is the best way to collect payment from people, I dont have a paypal account so do I just accept payment through mail and then ship out from there?

I would suggest setting up a Pay Pal account. Quick, easy and painless. You'll likely get more potential buyers that way as well. Otherwise you're limited to check or money orders for payment. Once you receive payment you would ship the item to your buyer. Have good communication between you and your buyer. Be clear about what you're selling and when you'll deliver and things should go smoothly for you.
