The 300 weighs less? Either way, go with the 300.

Can I assume that you're shopping for a "new" 621 and are thinking of buying/ordering the 300 vs the 250? Let's just be honest, if you can afford a new/newer 621 (either brand new or from the past year or so), you can afford the gas for either motor. The only concern I'd have is if you're gonna fish some tx's that limit hp to 250. Beyond that, they're both big motors fed by big gas tanks that will take a down payment to fill up this summer. I've got a 225 so I'm there with you.

There's always external considerations, but the majority of the time I say it's just like a trolling motor, "get the biggest that will fit and that you can afford." Comparing a 115 to a 300, yeah let's talk mileage and weight (but then we're talking different hulls entirely)...but 250 to 300...take the hp and never look back.
