You really should go through the floor and add fiberglass to the inside before sanding the outer 'patch'. Cut a hole big enough to get your hand through, you can but access ports that will cover it up when you are done. Make SURE you seal the floor end grain anywhere you cut it with silicone sealant and seal all fasteners.

To sand the outer patch, start with 600 grit and lightly bring the surface to level. Then use wet sanding paper, and when level and as nice as you want it, buff with polishing compound. If color matters to you, you will need to spray gel coat on the newly leveled surface, and somewhat complicated process, or use epoxy paint following all recommended prep procedures to a T.
